چکیده

To advance ecological civilization, developing sustainable, eco-friendly high-strength and ultra-tough alternatives to non-sustainable synthetic fibers, such as nylon, is crucial. This necessitates a deeply scientific understanding of the fundamental determinants fiber strength toughness, well overcoming engineering challenges for cost-effective, large-scale production high-performance silk fibers. Inspired by mechanical properties polyamide including nylon Kevlar, we employed CRISPR-Cas9-mediated gene editing successfully synthesize whole spider fibers from transgenic silkworms. These exhibited impressive tensile (1,299 MPa) toughness (319 MJ/m3), surpassing Kevlar’s 6-fold. Thus, they offer promising potential sustainable commercial Furthermore, our research provides valuable insights into essence strength, challenging conventional notion that these are contradictory. findings have significant implications guiding simultaneously possess high ultra-toughness.

Spider silk fibers spun from soluble recombinant silk produced in mammalian cells.

Spider silks are protein-based "biopolymer" filaments or threads secreted by specialized epithelial cells as concentrated soluble precursors of highly repetitive primary sequences. Spider dragline silk is a flexible, lightweight fiber of extraordinary strength and toughness comparable to that of synthetic high-performance fibers. We sought to "biomimic" the process of spider silk production by ...

Silkworms transformed with chimeric silkworm/spider silk genes spin composite silk fibers with improved mechanical properties.

The development of a spider silk-manufacturing process is of great interest. However, there are serious problems with natural manufacturing through spider farming, and standard recombinant protein production platforms have provided limited progress due to their inability to assemble spider silk proteins into fibers. Thus, we used piggyBac vectors to create transgenic silkworms encoding chimeric...

Silken toolkits: biomechanics of silk fibers spun by the orb web spider Argiope argentata (Fabricius 1775).

Orb-weaving spiders spin five fibrous silks from differentiated glands that contain unique sets of proteins. Despite diverse ecological functions, the mechanical properties of most of these silks are not well characterized. Here, we quantify the mechanical performance of this toolkit of silks for the silver garden spider Argiope argentata. Four silks exhibit viscoelastic behaviour typical of po...

Proteomic Evidence for Components of Spider Silk Synthesis from Black Widow Silk Glands and Fibers

Spider silk research has largely focused on spidroins, proteins that are the primary components of spider silk fibers. Although a number of spidroins have been characterized, other types of proteins associated with silk synthesis are virtually unknown. Previous analyses of tissue-specific RNA-seq libraries identified 647 predicted genes that were differentially expressed in silk glands of the W...

High-Toughness Silk Produced by a Transgenic Silkworm Expressing Spider (Araneus ventricosus) Dragline Silk Protein

Spider dragline silk is a natural fiber that has excellent tensile properties; however, it is difficult to produce artificially as a long, strong fiber. Here, the spider (Araneus ventricosus) dragline protein gene was cloned and a transgenic silkworm was generated, that expressed the fusion protein of the fibroin heavy chain and spider dragline protein in cocoon silk. The spider silk protein co...
